718-plex qPCR in single tube (1-shot optimization)

With a proven proof-of-concept that accommodates up to 718 primer pairs in a single reaction, DIMPLE drastically minimizes dimers and off-target interactions. This leads to a higher recovery yield, allowing for lower sample volumes without sacrificing sensitivity.

1000-plex 0-SHOT DIMPLE Designs

DIMPLE-designed primers work well with 0-shot optimization. (a) 1000-plex qPCR with 0-shot optimization shows effective amplification of 10 ng human gDNA over non-template control (NTC). When analyzed by NGS (b) 93.2% targets had at least 1 read, and 74.1% of targets were uniform within at least 0.2x median depth. (c) 100 different 10-plex qPCR sets of tarogets with 0-shot amplification. 97% show good specificity (NTC Ct>30), 98% show good sensitivity (10ng input human gDNA Ct<25), and 95% have both good sensitivity and specificity.
